Neuroprotection Effects of Ischemic Preconditioning in Posterior Circulation Ischemia Rat Model

Abstract
The aim of this study was to determine the neuroprotection effects of ischemic preconditioning (IPC) in vertebrobasilar ischemia induced by 4-vessel-occlusion (4VO) rat model. The Sprague-Dawley male rats weighting 230-300 g were randomly allocated to either the IPC-4VO group or 4VO group. The IPC-4VO group had an ischemic preconditioning three days before 4VO for posterior circulation ischemia. The ischemic preconditioning was induced by ligating both common carotid arteries for five minutes. For 4VO, both vertebral arteries were permanently occluded, and bilateral common carotid arteries were temporarily occluded for 20 minutes with a ligation. One hour later, the animals were sacrificed and subjected to immunohistochemical studies. The rats in the IPC-4VO group showed a significant decrease in the number of neurons showing c-fos protein expression in the medial vestibular nucleus compared to those in the 4VO group (20.8¡¾ 8.1 vs 36.8¡¾ 11.9, Wilcoxon rank test, P= 0.044). The ischemic preconditioning appears to have a neuroprotective effect on the vestibular system during posterior circulation ischemia.
